Descripción

PROKR2, another G protein-coupled receptor for prokineticins, shares functional overlap with PROKR1 but has distinct tissue distribution and physiological roles. It is highly expressed in the central nervous system, particularly in the hypothalamus and olfactory bulb, where it regulates circadian rhythms, neurogenesis, and reproductive function. PROKR2 couples to Gq and Gi/o proteins, stimulating phospholipase C (PLC) and MAPK pathways to regulate neuronal signaling, calcium homeostasis, and cellular responses. PROKR2 is crucial for hypothalamic-pituitary-gonadal axis function, influencing puberty onset and fertility. Mutations in PROKR2 are associated with Kallmann syndrome, characterized by delayed puberty and anosmia (loss of smell). PROKR2 also plays a role in thermoregulation, stress responses, and inflammation. Dysregulation of PROKR2 signaling contributes to reproductive disorders, sleep disturbances, and neuroinflammatory diseases. Targeting PROKR2 holds therapeutic promise for conditions such as infertility, sleep regulation disorders, and neurodevelopmental syndromes.
