Descripción
PLEKHA8, also known as TAPP1 (tandem PH-domain-containing protein 1), is a phosphoinositide-binding protein that interacts with PI(3,4)P2 to regulate intracellular trafficking and endosomal sorting processes. It functions in the recruitment of signaling complexes to membrane compartments, facilitating endocytosis, vesicle transport, and cellular polarity. PLEKHA8 is expressed in a variety of tissues, including epithelial cells, neurons, and immune cells, where it contributes to processes such as membrane dynamics, actin reorganization, and signal transduction. It plays an essential role in endosomal trafficking pathways, ensuring proper receptor recycling and signaling molecule localization. Dysregulation of PLEKHA8 has been implicated in cancer and neurodevelopmental disorders, where impaired membrane trafficking and signaling disrupt cell polarity and migration. Knockdown studies demonstrate defects in vesicular transport, reduced phosphoinositide binding, and altered cellular polarity, underscoring its critical role in intracellular trafficking and phosphoinositide-mediated signaling.

Cargo transport protein that is required for apical transport from the Golgi complex. Transports AQP2 from the trans-Golgi network(TGN) to sites of AQP2 phosphorylation. Mediates the non-vesicular transport of glucosylceramide(GlcCer) from the trans-Golgi network(TGN) to the plasma membrane and plays a pivotal role in the synthesis of complex glycosphingolipids. Binding of both phosphatidylinositol 4-phosphate(PIP) and ARF1 are essential for the GlcCer transfer ability. Also required for primary cilium formation, possibly by being involved in the transport of raft lipids to the apical membrane, and for membrane tubulation.
