Descripción
ABCB4, also known as MDR3, is expressed on the canalicular membrane of hepatocytes and plays a pivotal role in bile formation by mediating the ATP-dependent translocation of phosphatidylcholine into bile. Phosphatidylcholine protects bile ducts from the detergent effects of bile salts, ensuring proper bile composition and preventing bile-related liver damage. Mutations in ABCB4 are associated with progressive familial intrahepatic cholestasis type 3 (PFIC3), low phospholipid-associated cholelithiasis (LPAC), and other cholestatic liver diseases. Dysregulation of ABCB4 leads to bile duct injury, inflammation, and fibrosis, ultimately resulting in liver dysfunction. As a critical regulator of bile composition, ABCB4 is a significant focus in the study of liver health and diseases related to bile metabolism.
