PAX Interacting Protein 1 (PAXIP1) is a multifunctional nuclear protein that plays a critical role in DNA damage response and repair, particularly through its involvement in homologous recombination repair by recruiting key repair factors to sites of DNA double-strand breaks. It interacts with transcription factor Pax proteins, modulating gene expression essential for embryonic development, cell cycle regulation, and differentiation processes. PAXIP1 is also a core component of histone methyltransferase complexes, where it helps mediate H3K4 methylation, a crucial epigenetic modification linked to transcriptional activation and chromatin structure regulation. Its activity influences diverse biological processes, including immune system development, as it is necessary for effective immunoglobulin class switch recombination in B cells. Aberrations in PAXIP1 expression or function are associated with impaired DNA repair and genomic instability, contributing to tumorigenesis and progression in various cancers. PAXIP1 also serves as a scaffolding protein, facilitating interactions between DNA repair enzymes and chromatin remodelers, ensuring proper response to genotoxic stress. Its regulation is controlled through phosphorylation, enabling dynamic association with chromatin and response proteins, underscoring its role as a pivotal regulator of genome integrity and transcriptional modulation.
