Description

This gene is a member of the paired box (PAX) family of transcription factors. Members of this gene family typically contain a paired box domain, an octapeptide, and a paired-type homeodomain. These genes play critical roles during fetal development and cancer growth. The paired box 4 gene is involved in pancreatic islet development and mouse studies have demonstrated a role for this gene in differentiation of insulin-producing beta cells.

Descripción

PAX4 is a transcription factor involved in the development and differentiation of pancreatic β-cells, essential for insulin production. PAX4 plays a critical role during embryonic development by repressing the α-cell lineage and promoting the specification of endocrine progenitor cells into β-cells within the pancreas. It directly regulates genes involved in β-cell identity, proliferation, and survival. Mutations in PAX4 are associated with monogenic forms of diabetes, such as maturity-onset diabetes of the young (MODY) and other metabolic disorders due to insufficient insulin production. PAX4 expression is tightly regulated, and its overexpression can influence β-cell regeneration, offering therapeutic potential for diabetes treatment. Knockout models show impaired β-cell development, hyperglycemia, and defective glucose homeostasis, underscoring PAX4's pivotal role in pancreatic endocrine development, insulin regulation, and diabetes pathology.
