Nuclear receptor subfamily 5 group A member 1 (NR5A1), also known as SF-1 (steroidogenic factor-1), is a nuclear receptor critical for regulating the development of the gonads, adrenal glands, and other steroidogenic tissues NR5A1 is essential for the transcription of genes involved in steroid hormone synthesis, including the biosynthesis of cortisol, aldosterone, and sex hormones It plays a pivotal role in the differentiation of gonadal and adrenal cells and the regulation of the hypothalamic-pituitary-gonadal axis, which controls reproductive function and hormone production In addition to its role in hormone regulation, NR5A1 is involved in the regulation of lipid metabolism and energy homeostasis Mutations in NR5A1 can lead to disorders such as congenital adrenal hyperplasia and disorders of sexual development, as well as infertility and adrenal insufficiency Dysregulation of NR5A1 is also linked to certain cancers, including gonadal tumors, highlighting its role as a key regulator of both reproductive and metabolic health

Binds to the Ad4 site found in the promoter region of steroidogenic P450 genes such as CYP11A, CYP11B and CYP21B.
