Descripción
NPFFR1, also known as GPR147, is a G protein-coupled receptor activated by neuropeptide FF (NPFF), a member of the RFamide peptide family. NPFFR1 is predominantly expressed in the central nervous system (CNS), particularly in regions involved in pain modulation, opioid signaling, and neuroendocrine regulation. Activation of NPFFR1 couples to Gi/o proteins, inhibiting adenylate cyclase and reducing cAMP levels, leading to the modulation of synaptic transmission and neuronal excitability. NPFFR1 plays a critical role in modulating opioid analgesia, acting as an anti-opioid receptor to counteract the effects of endogenous and exogenous opioids, including morphine tolerance and dependence. Additionally, NPFFR1 is involved in regulating stress responses, cardiovascular function, and neuroendocrine signaling. Dysregulation of NPFFR1 has been associated with chronic pain, opioid tolerance, and stress-related disorders. NPFFR1 antagonists are being investigated as potential therapies for enhancing opioid analgesia and reducing tolerance, while its broader roles in stress and cardiovascular regulation remain under exploration.
