Background

Neurofibromin 1 (NF1) is a tumor suppressor protein that plays a critical role in regulating cell growth, differentiation, and survival NF1 functions primarily as a negative regulator of the RAS signaling pathway, which is involved in cellular processes such as proliferation, differentiation, and survival By hydrolyzing GTP to GDP, neurofibromin inactivates RAS, thus preventing uncontrolled cell growth and the formation of tumors Neurofibromin is expressed in various tissues, including the nervous system, where it helps maintain normal neurodevelopment and prevent tumor formation Disruptions or mutations in the NF1 gene lead to neurofibromatosis type 1 (NF1), a genetic disorder characterized by the development of benign tumors, called neurofibromas, along nerves Additionally, NF1 mutations are associated with an increased risk of malignant tumors, learning disabilities, and other neurological symptoms The role of NF1 in tumor suppression and cellular signaling makes it an important target for therapeutic interventions aimed at managing neurofibromatosis and related cancers
