SIRT1 is a NAD+-dependent deacetylase and member of the sirtuin family that regulates cellular processes including gene expression, metabolism, DNA repair, and aging by removing acetyl groups from target proteins SIRT1 is highly expressed in various tissues, including the liver, brain, and skeletal muscle, where it modulates the activity of transcription factors such as p53, FOXO, NF-κB, and PPARγ coactivator-1α (PGC-1α) SIRT1 promotes cellular survival during stress by enhancing DNA repair, suppressing apoptosis, and regulating energy metabolism through mitochondrial biogenesis and fatty acid oxidation Activation of SIRT1 is associated with increased longevity and protection against age-related diseases such as type 2 diabetes, cardiovascular disease, neurodegeneration, and cancer Compounds like resveratrol act as SIRT1 activators to improve metabolic health by enhancing insulin sensitivity and mitochondrial function SIRT1 also inhibits inflammation by deacetylating NF-κB and reducing pro-inflammatory cytokine production, making it a key target for therapies in metabolic, inflammatory, and age-associated disorders

This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class I of the sirtuin family. Alternative splicing results in multiple transcript variants.
