Descripción
DENN domain containing 2B (DENND2B) is a guanine nucleotide exchange factor (GEF) that specifically activates Rab GTPases, particularly Rab9, facilitating vesicular trafficking between late endosomes and the trans-Golgi network. Through its DENN domain, DENND2B promotes the GDP-GTP exchange on Rab9, thereby enabling the transport of vesicles containing enzymes, receptors, and other cargo necessary for lysosomal and Golgi function. DENND2B plays a crucial role in maintaining the integrity of lysosomal and endosomal compartments and is involved in processes such as protein recycling and cellular nutrient homeostasis. Its expression is widespread, with enhanced activity in tissues requiring high levels of vesicle transport, such as neural and immune cells. Dysregulation of DENND2B has been implicated in neurodegenerative and metabolic disorders, where defects in vesicle trafficking lead to the accumulation of undegraded substrates and disrupted cellular signaling.
