Descripción
SH2D2A, also known as SCOP (SH2 domain-containing phosphatase), is an SH2 domain-containing adaptor protein that regulates T-cell receptor (TCR) and cytokine receptor signaling pathways. It is primarily expressed in T cells and other hematopoietic cells, where it modulates immune cell activation, proliferation, and cytokine production. SH2D2A functions by binding to phosphorylated receptors and signaling intermediates, linking TCR activation to downstream pathways like NF-κB, MAPK, and PI3K/Akt. It plays an important role in T-cell activation, survival, and differentiation, particularly during immune responses. Dysregulation of SH2D2A is associated with immune dysfunction, autoimmune diseases, and impaired T-cell responses. Knockout studies demonstrate defects in T-cell activation, reduced cytokine production, and impaired antigen-specific responses, underscoring its role as a critical regulator of TCR signaling and adaptive immunity.
