Ragulator complex protein LAMTOR2 is a key component of the Ragulator complex, which anchors Rag GTPases to the lysosomal membrane, facilitating mTORC1 activation in response to amino acid availability LAMTOR2 works alongside other Ragulator subunits to enable the recruitment and activation of mTORC1, a master regulator of cellular metabolism, growth, and autophagy LAMTOR2 also plays a role in endosomal-lysosomal trafficking and cellular nutrient sensing Studies show that LAMTOR2 is critical for immune cell function, as its loss impairs mTORC1 signaling and compromises T-cell activation and proliferation Dysregulation of LAMTOR2 expression has been implicated in cancer, where disrupted mTOR signaling promotes tumor cell survival and metabolic reprogramming Additionally, defects in LAMTOR2 are associated with lysosomal storage disorders and developmental abnormalities, emphasizing its role in cellular homeostasis, autophagy, and organismal development
