Descripción
Proteinase-Activated Receptor 3 (F2RL2), also known as PAR3, is another member of the PAR family of receptors. It is primarily expressed in platelets, endothelial cells, and the gastrointestinal tract. F2RL2 is activated by thrombin, which cleaves the receptor to reveal a tethered ligand that activates intracellular signaling. Unlike other PARs, F2RL2 acts in concert with F2RL1 (PAR2) to modulate thrombin-induced platelet activation and coagulation. F2RL2 couples to Gi and Gq proteins, triggering a variety of signaling events such as platelet aggregation, smooth muscle contraction, and endothelial cell activation. Its primary role is in regulating hemostasis and thrombus formation, particularly during tissue injury or inflammation. Dysregulation of F2RL2 signaling can contribute to thrombosis, atherosclerosis, and ischemic conditions. Targeting F2RL2 offers therapeutic potential in cardiovascular diseases, particularly those involving abnormal platelet activation and clot formation, by preventing excessive thrombosis without impairing normal wound healing.
