NMUR1 is a G protein-coupled receptor activated by neuromedin U (NMU), a neuropeptide involved in regulating appetite, energy homeostasis, and inflammation. NMUR1 is primarily expressed in peripheral tissues, including the gastrointestinal tract, lungs, and immune cells, where it mediates NMU’s effects on smooth muscle contraction, inflammation, and immune responses. Upon activation, NMUR1 couples to Gq/11 proteins, leading to phospholipase C (PLC) activation, calcium mobilization, and smooth muscle contraction. NMUR1 plays a role in regulating gut motility, bronchoconstriction, and inflammatory responses, particularly in allergic conditions such as asthma and inflammatory bowel disease (IBD). Dysregulation of NMUR1 signaling contributes to inflammatory and metabolic disorders, where it amplifies immune responses and modulates energy expenditure. NMUR1 antagonists are being investigated for their potential to treat inflammatory diseases, asthma, and obesity, as NMU signaling through NMUR1 influences inflammation, energy balance, and smooth muscle activity.
