Background

GRM4, or mGluR4, belongs to the Group III metabotropic glutamate receptors and primarily couples to Gi/o proteins, inhibiting adenylate cyclase and reducing cyclic AMP (cAMP) levels. GRM4 is predominantly expressed in the cerebellum, basal ganglia, and thalamus, where it functions as a presynaptic receptor to suppress excessive glutamate release. It plays a critical role in regulating synaptic plasticity, motor coordination, and neuronal excitability. GRM4 activation has neuroprotective effects, reducing excitotoxicity and inflammation in neurological conditions.
