GRM4, or mGluR4, belongs to the Group III metabotropic glutamate receptors and primarily couples to Gi/o proteins, inhibiting adenylate cyclase and reducing cyclic AMP (cAMP) levels. GRM4 is predominantly expressed in the cerebellum, basal ganglia, and thalamus, where it functions as a presynaptic receptor to suppress excessive glutamate release. It plays a critical role in regulating synaptic plasticity, motor coordination, and neuronal excitability. GRM4 activation has neuroprotective effects, reducing excitotoxicity and inflammation in neurological conditions. Dysregulation of GRM4 has been associated with Parkinson’s disease, anxiety, and chronic pain, where impaired glutamate signaling contributes to disease pathology. GRM4 agonists are being investigated as therapeutic agents for Parkinson’s disease, as they modulate basal ganglia circuitry and improve motor symptoms. Additionally, GRM4’s role in neuroprotection and reducing inflammation makes it a promising target for treating neurodegenerative diseases, chronic pain, and psychiatric disorders.

GRM4 Antibody is a Rabbit Polyclonal antibody against GRM4. L-glutamate is the major excitatory neurotransmitter in the central nervous system and activates both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. The metabotropic glutamate receptors are a family of G protein-coupled receptors, that have been divided into 3 groups on the basis of sequence homology, putative signal transduction mechanisms, and pharmacologic properties. Group I includes GRM1 and GRM5 and these receptors have been shown to activate phospholipase C. Group II includes GRM2 and GRM3 while Group III includes GRM4, GRM6, GRM7 and GRM8. Group II and III receptors are linked to the inhibition of the cyclic AMP cascade but differ in their agonist selectivities. Several transcript variants encoding different isoforms have been found for this gene.
