Background

GRM4, or mGluR4, belongs to the Group III metabotropic glutamate receptors and primarily couples to Gi/o proteins, inhibiting adenylate cyclase and reducing cyclic AMP (cAMP) levels. GRM4 is predominantly expressed in the cerebellum, basal ganglia, and thalamus, where it functions as a presynaptic receptor to suppress excessive glutamate release. It plays a critical role in regulating synaptic plasticity, motor coordination, and neuronal excitability. GRM4 activation has neuroprotective effects, reducing excitotoxicity and inflammation in neurological conditions. Dysregulation of GRM4 has been associated with Parkinson’s disease, anxiety, and chronic pain, where impaired glutamate signaling contributes to disease pathology. GRM4 agonists are being investigated as therapeutic agents for Parkinson’s disease, as they modulate basal ganglia circuitry and improve motor symptoms.
