Descripción
LPAR5 is a G protein-coupled receptor activated by lysophosphatidic acid (LPA), primarily coupling to G12/13 and Gq proteins. It is expressed in the gastrointestinal tract, immune cells, and sensory neurons, where it regulates cellular responses such as migration, cytoskeletal reorganization, and inflammatory signaling. LPAR5 plays a significant role in immune modulation and inflammatory responses by mediating immune cell trafficking and cytokine release. In the gastrointestinal tract, LPAR5 regulates intestinal epithelial cell homeostasis and motility, making it relevant in gut inflammation and tissue repair. Additionally, LPAR5 is implicated in neuropathic pain, where its activation sensitizes sensory neurons, contributing to pain perception. Dysregulation of LPAR5 signaling has been linked to inflammatory diseases, neuropathic pain, and cancer progression, where it enhances tumor cell migration and survival. LPAR5 antagonists are under investigation for therapeutic use in conditions such as inflammatory bowel disease, neuropathic pain, and cancer metastasis, highlighting its potential as a target for immune and inflammatory regulation.
