LRRC51 is a key component of the CATSPER complex, a calcium channel specifically located in the flagellum of sperm cells that regulates sperm motility and hyperactivation, essential for fertilization. It contains leucine-rich repeats (LRRs) that mediate protein-protein interactions, enabling it to assemble and stabilize the CATSPER channel alongside other subunits like CATSPER1-4 and LRRC52. LRRC51 plays a critical role in mediating calcium influx into sperm in response to physiological cues, allowing flagellar beating and hyperactivation during capacitation. Mutations or dysfunction of LRRC51 result in impaired sperm motility and male infertility, as the CATSPER complex is non-functional without its proper assembly. LRRC51 is exclusively expressed in the testes, specifically in mature sperm cells, underscoring its role in reproductive biology. Knockout models exhibit severely compromised sperm motility, defects in capacitation, and infertility, emphasizing its essential function in sperm function and male fertility.
