Descripción
GRB7 is an adaptor protein that interacts with receptor tyrosine kinases (RTKs) like HER2, VEGFR, and IGF1R, linking them to downstream signaling pathways such as Ras/MAPK, PI3K/Akt, and focal adhesion kinase (FAK). GRB7 contains SH2, PH, and proline-rich domains, enabling its localization to membranes and interactions with signaling molecules involved in cell migration, adhesion, and proliferation. GRB7 is highly expressed in cancers, particularly HER2-positive breast cancer, ovarian cancer, and esophageal carcinoma, where it promotes tumor progression, metastasis, and angiogenesis. It also regulates integrin-mediated signaling pathways, playing roles in cell spreading and cytoskeletal organization. GRB7 dysregulation is associated with poor clinical outcomes in cancer patients due to its role in enhancing cell motility and survival. Knockdown studies show reduced tumor cell invasion, highlighting its potential as a therapeutic target in metastatic cancers.
