C-C Chemokine Receptor Type 6 (CCR6) is a G-protein-coupled receptor expressed on subsets of B cells, T-helper 17 (Th17) cells, regulatory T cells (Tregs), dendritic cells, and memory T cells. CCR6 binds its ligand CCL20 (MIP-3α) to regulate immune cell migration to sites of inflammation, infection, and mucosal tissues. It plays a pivotal role in mucosal immunity, promoting the trafficking of lymphocytes and dendritic cells to the gut-associated lymphoid tissue (GALT), where it contributes to host defense and immune homeostasis. CCR6 is essential for Th17 cell recruitment, which is critical for combating extracellular bacteria and fungi but can also drive autoimmune diseases like psoriasis, rheumatoid arthritis, and inflammatory bowel disease (IBD) when dysregulated. CCR6 also facilitates immune tolerance through its expression on Tregs, balancing pro-inflammatory and anti-inflammatory responses. In cancer, CCR6 is associated with tumor progression and metastasis, particularly in colorectal cancer and hepatocellular carcinoma, by mediating immune cell infiltration and fostering a tumor-permissive microenvironment. The CCR6-CCL20 axis has also been implicated in HIV infection, as CCR6+ T cells are preferential targets for viral replication. Targeting CCR6 and its signaling pathways holds therapeutic promise for autoimmune diseases, infectious diseases, and cancer immunotherapy. Ongoing research continues to elucidate its complex roles in immunity, inflammation, and disease pathology.
