Descripción
AKAP9 is a large scaffold protein localized to centrosomes, Golgi apparatus, and microtubules, where it organizes PKA and other signaling complexes involved in cell cycle regulation, microtubule dynamics, and centrosome cohesion. AKAP9 interacts with proteins such as PKA, phosphatases, and cyclin-dependent kinases (CDKs), regulating mitotic spindle formation, chromosome segregation, and cytokinesis. It is essential for centrosome duplication and microtubule nucleation during interphase and mitosis, ensuring proper cell division. AKAP9 is also involved in Golgi apparatus positioning and trafficking, where it interacts with motor proteins and small GTPases to coordinate intracellular transport. Mutations in AKAP9 are associated with ciliopathies, neurodevelopmental disorders, and cancer due to disrupted centrosome function and mitotic defects. AKAP9 has also been shown to regulate cardiac excitation-contraction coupling by anchoring PKA to ion channels and calcium-handling proteins. Its role in both structural and signaling organization makes it a key regulator of cell division, polarity, and organelle function.
