LRRC4, also known as netrin-G ligand 2 (NGL-2), is a leucine-rich repeat-containing protein involved in synaptic organization, neuronal development, and cell signaling. It serves as a postsynaptic adhesion molecule that interacts with netrin-G2, facilitating synapse formation, maintenance, and stability in the central nervous system. LRRC4 is highly expressed in the brain, particularly in regions associated with synaptic plasticity, learning, and memory. It regulates axon guidance, dendritic development, and excitatory synaptic transmission by interacting with cytoskeletal proteins and signaling molecules. Dysregulation or mutations in LRRC4 are associated with neurological disorders, including autism spectrum disorders (ASD) and intellectual disabilities, where impaired synaptic organization disrupts neuronal connectivity. LRRC4 also exhibits tumor-suppressive properties in gliomas by inhibiting cell proliferation, migration, and survival pathways. Knockout studies reveal impaired synaptic function, altered dendritic morphology, and reduced neuronal connectivity, highlighting its essential role in neural circuit development, synaptic organization, and brain function.
