Descripción

CXCR2 is a G protein-coupled receptor that binds IL-8 and other CXC chemokines like Groα, Groβ, and NAP-2. It is mainly expressed on neutrophils, endothelial cells, and monocytes, mediating chemotaxis, neutrophil activation, and cytokine release during inflammation. CXCR2 signaling activates pathways such as PI3K, MAPK, and NF-κB, while its rapid desensitization and internalization regulate signal duration. CXCR2 is critical for immune surveillance and inflammation but also contributes to diseases like COPD, asthma, and rheumatoid arthritis due to excessive neutrophil infiltration. In cancer, CXCR2 promotes tumor progression, angiogenesis, and metastasis by recruiting myeloid-derived suppressor cells (MDSCs) and endothelial cells to the tumor microenvironment, enhancing immunosuppression and vascularization. CXCR2 inhibition reduces tumor growth and inflammation, highlighting its therapeutic potential. It also regulates leukocyte trafficking and tissue repair but contributes to pathologies like cystic fibrosis and inflammatory bowel disease (IBD) when dysregulated. Overall, CXCR2 is key for immune cell recruitment and inflammation, balancing host defense with disease progression.
