The insulin-like growth factor 1 receptor (IGF1R) is a transmembrane receptor that plays a crucial role in cellular growth, survival, and metabolism. It is activated primarily by insulin-like growth factor 1 (IGF-1), a hormone structurally similar to insulin but with distinct biological functions. IGF1R is a member of the receptor tyrosine kinase (RTK) family, which includes receptors like the insulin receptor (INSR). IGF1R signaling is essential for normal development and is involved in many physiological processes, such as cell differentiation, proliferation, and apoptosis inhibition. This receptor is also significant in pathological states, especially cancer, where it can drive tumor growth and metastasis through enhanced cell survival and proliferation pathways.

IGF1R, also named as CD221, is a receptor tyrosine kinase which mediates actions of insulin-like growth factor 1 (IGF1). IGF1R binds IGF1 with high affinity and IGF2 and insulin (INS) with a lower affinity, the activated IGF1R is involved in cell growth and survival control. It is crucial for tumor transformation and survival of malignant cell. IGF1R is a heterotetrameric receptor, composed of two alpha subunits (130 kDa each) and two beta subunits (~98 kDa each) (PMID: 6279656). Both the alpha and beta subunits are synthesized from a single mRNA precursor. The precursor is then glycosylated, proteolytically cleaved, and crosslinked by cysteine bonds.
