Background
UBX domain protein 2A (UBXN2A) is a p97/VCP-interacting protein containing a UBX domain that regulates protein degradation pathways including endoplasmic reticulum-associated degradation (ERAD) and ubiquitin-proteasome systems UBXN2A acts as a cofactor for p97, facilitating the extraction and degradation of misfolded proteins and maintaining proteostasis under cellular stress conditions It also modulates autophagy by linking p97 to lysosomal pathways, ensuring the clearance of protein aggregates Dysregulation of UBXN2A expression has been linked to cancer progression, where its involvement in protein turnover supports tumor cell survival, metastasis, and resistance to therapy Emerging studies show that UBXN2A is critical for cellular stress responses, DNA repair, and apoptosis regulation in response to proteotoxic insults UBXN2A’s role in proteasome-mediated degradation and its influence on cellular homeostasis underscore its importance in neurodegeneration and cancer-related proteostasis dysfunction
