| Assay Type | Activity: Active Biological Activity: EC50: 0.412 µg/ml A functional binding ELISA assay was conducted to detect the interaction of recombinant Human CD80 with recombinant human CD86. Briefly, serial dilutions of recombinant rat Human T-lymphocyte activation antigen CD80 / B7-1 (CD80) Protein were carried out in PBS, 0.01 % BSA (pH 7.4). Duplicate samples of 100 µl were transferred to microtiter wells pre-coated with CD86 and incubated for 1h at 37 °C. Wells were washed with PBST 3 times and incubated with Streptavidin-HRP for 40 minutes, then aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 °C. Finally, add 50 µl stop solution to the wells and read at 450 nm immediately. Many proteins are synthesised as pro-proteins, and then cleaved to give the active form. 5. Polymerisation of the target protein. Dimerisation, multimerisation etc. will increase the band size observed. |
