Background

SH2D6 is an adaptor protein containing an SH2 domain, functioning as a mediator of signal transduction pathways downstream of activated cytokine and growth factor receptors. It participates in pathways such as JAK/STAT and MAPK, facilitating signal relay to regulate cell proliferation, differentiation, and survival. SH2D6 is expressed predominantly in hematopoietic and immune cells, where it plays roles in cytokine signaling, lymphocyte development, and immune cell activation. It interacts with signaling proteins via its SH2 domain, bridging upstream receptors to downstream effectors during immune responses. Dysregulation of SH2D6 has been linked to immune dysfunction and inflammatory disorders due to impaired cytokine signaling regulation. Knockout models show defects in lymphocyte activation, reduced cytokine production, and abnormal immune responses, emphasizing its importance in maintaining immune signaling and homeostasis.
