Descripción
Proteinase-Activated Receptor 4 (F2RL3), or PAR4, is activated by thrombin and other serine proteases, including trypsin. It is primarily expressed in platelets, endothelial cells, and various immune cells. F2RL3 plays a crucial role in platelet aggregation, clot formation, and wound healing. Upon activation, PAR4 signals through Gi proteins to modulate cellular responses such as platelet activation, granule release, and thrombus formation. PAR4 is a key mediator of thrombin-induced platelet responses and works in concert with other PARs (like PAR1 and PAR3) to regulate the coagulation cascade. Dysregulation of F2RL3 signaling has been associated with thrombotic disorders, such as deep vein thrombosis and stroke. PAR4 antagonists are being investigated for their potential to prevent thrombosis, especially in cardiovascular diseases where clot formation is a major contributor to disease progression. Additionally, F2RL3’s role in immune cell activation and inflammation suggests it could be a target for managing inflammatory diseases and autoimmune disorders, as well as regulating immune responses during infection.
