Background
PRLHR, also known as the G protein-coupled receptor for prolactin-releasing peptide (PrRP), regulates energy homeostasis, stress responses, and endocrine functions. PRLHR is predominantly expressed in the hypothalamus and brainstem, where it modulates prolactin release, appetite suppression, and metabolic processes. PRLHR couples to Gq proteins, activating phospholipase C (PLC), increasing intracellular calcium, and stimulating downstream pathways involved in neurotransmission and hormone regulation. Beyond prolactin release, PRLHR plays a role in regulating feeding behavior by acting as an anorexigenic signal that reduces food intake and promotes energy expenditure. PRLHR is also implicated in cardiovascular regulation and stress-related behaviors through its actions in the autonomic nervous system. Dysregulation of PRLHR has been linked to obesity, metabolic syndrome, and stress disorders. Therapeutic targeting of PRLHR is being explored for obesity management and metabolic disorders, as its activation can enhance satiety and improve energy balance while modulating neuroendocrine responses.
