Ragulator complex protein LAMTOR3 is an integral subunit of the lysosomal Ragulator complex that functions as a critical activator of the mTORC1 pathway in response to amino acid and nutrient signaling LAMTOR3 is required for the localization of Rag GTPases to the lysosomal surface, which facilitates the recruitment of mTORC1 and its activation upon nutrient availability Beyond its role in nutrient sensing, LAMTOR3 regulates lysosomal positioning and intracellular trafficking processes Dysregulation of LAMTOR3 disrupts mTOR signaling and has been linked to cellular growth defects, metabolic disorders, and tumor progression Studies highlight its role in maintaining energy balance and cell survival under stress conditions LAMTOR3 also participates in immune cell responses, as mTORC1 signaling is vital for T-cell activation and differentiation Its involvement in nutrient sensing and cellular homeostasis underscores its importance in both normal physiology and disease pathogenesis
