| Assay Type | Activity: Active Biological Activity: Interleukin 17C (IL17C) is a T cell-derived cytokine which has high sequence homology with Interleukin 17 (IL17) and may play a crucial role in innate immunity of the epithelium. IL17C binds to Interleukin 17 receptor A (IL17RA), thus a binding ELISA assay was conducted to detect the interaction of recombinant human IL17C and recombinant human IL17RA. Briefly, IL17C was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 µl were then transferred to IL17RA-coated microplate wells and incubated for 2 h at 37°C. Wells were washed with PBST and incubated for 1 h with anti-IL17C polyclonal antibody, then aspirated and washed 3 times. After incubation with HRP-conjugated secondary antibody, wells were aspirated and washed 3 times. TMB substrate solution was added and wells were incubated for 15-25 minutes at 37 °C. Finally, 50 µl stop solution was added to the wells and the absorbance was read at 450 nm immediately. Sequence Fragment: MG + His19-Val197 + LEHHHHHH Tag: C-terminal His tag |

| Observed MW | Calculated MW: 21.0 kDa Observed MW (SDS-PAGE): 25 kDa Possible reasons why the actual band size differs from the predicted band size: 1. Splice variants. Alternative splicing may create different sized proteins from the same gene. 2. Relative charge. The composition of amino acids may affect the charge of the protein. 3. Post-translational modification. Phosphorylation, glycoslyation, methylation etc. may affect the band size. 4. Post-translational cleavage. Many proteins are synthesised as pro-proteins, and then cleaved to give the active form. 5. Polymerisation of the target protein. Dimerisation, multimerisation etc. will increase the band size observed. |

| Note | THIS PRODUCT IS FOR RESEARCH USE ONLY. NOT FOR USE IN DIAGNOSTIC, THERAPEUTIC OR COSMETIC PROCEDURES. NOT FOR HUMAN OR ANIMAL CONSUMPTION. To keep the original salt concentration, we recommend reconstituting to the original concentration prior to lyophilization (see Concentration) in ddH2O. If a lower concentration is required, dilute in 20 mM Tris, 150 mM NaCl, pH 8.0. If a higher concentration is required, the product can be reconstituted directly in 20 mM Tris, 150 mM NaCl, pH 8.0, though please note that this will change the overall salt concentration. The stock concentration should be between 0.1-1.0 mg/ml. Do not vortex. Concentration: Prior to lyophilization: 100 µg/ml |
Descripción
Interleukin 17C (IL17C) is a member of the IL-17 cytokine family, which is heavily involved in immune responses, particularly in inflammatory and autoimmune processes. The IL-17 family includes six members: IL-17A, IL-17B, IL-17C, IL-17D, IL-17E (IL-25), and IL-17F, all of which contribute in varying ways to host defense, primarily by inducing inflammatory responses. IL-17C plays a distinct role in mucosal immunity, primarily by acting as a mediator of the host defense against bacterial pathogens, particularly at epithelial barriers. IL-17C has drawn interest because of its autocrine and paracrine signaling capabilities, meaning that it can act both on the cells that produce it (autocrine signaling) and on neighboring cells (paracrine signaling). In contrast to IL-17A and IL-17F, which are secreted by Th17 cells, IL-17C is predominantly produced by epithelial cells. Its role in enhancing the immune response to infection makes it critical for barrier defense in tissues like the skin, lungs, intestines, and other mucosal surfaces
