Descripción

ALOXE3 is a member of the lipoxygenase family that functions as an epidermal lipoxygenase critical for skin barrier formation and maintenance. Unlike other lipoxygenases, ALOXE3 acts as a hydroperoxide isomerase, converting epoxyalcohol intermediates into key lipid mediators required for the assembly of the cornified lipid envelope in the epidermis. ALOXE3 is specifically expressed in the skin and is essential for proper lipid metabolism during keratinocyte differentiation. Mutations in the ALOXE3 gene are linked to autosomal recessive congenital ichthyosis (ARCI), a disorder characterized by dry, scaly skin due to defective epidermal lipid processing and barrier dysfunction. In addition to its role in skin biology, ALOXE3 may contribute to anti-inflammatory pathways by regulating lipid signaling in epithelial tissues. Emerging research suggests that ALOXE3 dysfunction may influence wound healing, oxidative stress responses, and epithelial tissue repair. Its unique enzymatic activity makes it essential for skin homeostasis and a potential therapeutic target for treating barrier-related skin diseases.

ALOXE3 is a member of the lipoxygenase family, which are catabolized by arachidonic acid-derived compounds. The encoded enzyme is a hydroperoxide isomerase that synthesizes a unique type of epoxy alcohol (8R-hydroxy-11R, 12R-epoxyeicosa-5Z, 9E, 14Z-trienoic acid) from 12R-hydroperoxyeicosatetraenoic acid (12R-HPETE). This epoxy alcohol can activate the the nuclear receptor peroxisome proliferator-activated receptor alpha (PPARalpha), which is implicated in epidermal differentiation. Loss of function of the enzyme encoded by this gene results in ichthyosis, implicating the function of this gene in the differentiation of human skin.
