Background
GAB3 is an adaptor protein structurally related to GAB1 and GAB2, functioning downstream of receptor tyrosine kinases to activate pathways like PI3K/Akt and MAPK/ERK. It is primarily expressed in hematopoietic cells and immune tissues, where it regulates cell survival, proliferation, and differentiation. GAB3 supports B-cell receptor (BCR) and cytokine-mediated signaling, playing a role in B-cell development and macrophage activation. Its dysregulation is associated with altered immune responses and hematopoietic disorders. GAB3 interacts with SH2-containing proteins to modulate signaling networks involved in immune cell function. Knockout studies suggest a role in regulating inflammatory responses and immune homeostasis, particularly in myeloid and lymphoid lineages.
