FNIP1 is a critical binding partner of folliculin (FCLN) and plays a key role in energy sensing and metabolic regulation through its interactions with AMPK and mTOR signaling pathways. FNIP1 acts as a molecular scaffold, facilitating the assembly of the FCLN-FNIP complex on lysosomal membranes, which is necessary for proper mTORC1 activation in response to nutrient and energy cues. FNIP1 also interacts with the Hsp90 chaperone system, regulating the stability and functionality of FCLN. Loss of FNIP1 impairs AMPK signaling, disrupting cellular energy homeostasis and leading to metabolic dysregulation. Studies in mouse models have shown that FNIP1 is essential for B-cell development, suggesting a broader role in lymphocyte function and immune responses. FNIP1 expression is highest in metabolically active tissues, including the heart, skeletal muscle, and kidney. Its role in coordinating FCLN function and metabolic signaling positions FNIP1 as a key regulator of cellular growth and proliferation, and its dysregulation is implicated in oncogenic and metabolic disorders.
