Descripción
DPH3 plays a supporting but essential role in diphthamide biosynthesis by stabilizing the DPH1-DPH2 complex and facilitating the transfer of intermediate molecules during the catalytic process. It functions as a small zinc-binding protein and is thought to act as an electron donor, aiding the reductive chemistry required for diphthamide formation. DPH3 mutations or downregulation lead to resistance to diphtheria toxin and defects in translational elongation due to impaired diphthamide synthesis. Unlike DPH1 and DPH2, DPH3 has a broader cellular role, including involvement in redox homeostasis and regulation of oxidative stress responses. DPH3 is also implicated in protein-protein interactions within the diphthamide biosynthesis complex, ensuring the proper assembly and function of the enzymatic machinery. Expression studies show that DPH3 is widely distributed across eukaryotic tissues, particularly in metabolically active cells. Loss-of-function mutations in DPH3 are associated with reduced cellular fitness and increased vulnerability to environmental stressors, further emphasizing its auxiliary but indispensable role in diphthamide synthesis.
