Descripción

DENN domain containing 5B (DENND5B) is a closely related paralog of DENND5A, acting as a guanine nucleotide exchange factor (GEF) for Rab GTPases and contributing to vesicular trafficking and lysosomal transport. DENND5B is particularly involved in activating Rab39, facilitating vesicle tethering, sorting, and fusion between late endosomes and lysosomes. It plays a key role in recycling membrane-bound receptors and transporters, ensuring efficient cellular signaling and nutrient uptake. DENND5B is also linked to autophagic processes, supporting the clearance of damaged cellular components and maintaining lysosomal integrity. Its expression is widespread, but it is highly active in tissues such as the brain and liver, which depend heavily on efficient vesicular transport systems. Dysregulation of DENND5B has been implicated in neurodegenerative diseases and metabolic syndromes, emphasizing its critical role in sustaining cellular proteostasis and responding to metabolic and environmental stress. Its close functional relationship with DENND5A highlights the importance of this subgroup in vesicular transport and cellular health.
