Descripción
DENN domain containing 4B (DENND4B) is another member of the DENN family that acts as a guanine nucleotide exchange factor (GEF) for Rab10, closely related in function to DENND4A. DENND4B regulates intracellular trafficking, membrane remodeling, and endocytic recycling by promoting Rab10 activation. It plays a particularly crucial role in polarized transport in epithelial cells, ensuring the targeted delivery of vesicle cargo to specific membrane domains. In metabolic tissues, DENND4B contributes to the recycling of glucose transporters, enhancing glucose uptake and maintaining cellular metabolic homeostasis. Its regulatory influence extends to signaling pathways linked to receptor recycling and secretion. DENND4B is expressed in a wide range of tissues, with high activity in metabolically active cells such as adipocytes and hepatocytes, which are heavily reliant on efficient vesicle transport systems. Dysregulation of DENND4B has been implicated in metabolic diseases, such as diabetes, as well as in defects in membrane trafficking pathways, further emphasizing its critical role in sustaining cellular function, signaling, and metabolic adaptability.
