Descripción
DOCK5, structurally similar to DOCK1, acts as a GEF for Rac1 and Cdc42, regulating actin polymerization during cell migration and bone resorption. It is essential for osteoclast function, promoting cytoskeletal organization and podosome formation. DOCK5 is expressed in skeletal and immune tissues, playing roles in osteoclast-mediated bone remodeling and macrophage migration. Dysregulation leads to bone diseases like osteoporosis due to impaired bone resorption. DOCK5 interacts with the DOCK–ELMO complex to mediate Rac1 activation. In cancer, DOCK5 enhances tumor cell motility and metastasis. Mouse models lacking DOCK5 exhibit reduced osteoclast activity and defects in immune cell migration.
