DDB1 and CUL4 associated factor 7 (DCAF7), also known as WDR68, functions as a substrate receptor within the CUL4-DDB1 E3 ubiquitin ligase complex and plays a critical role in protein ubiquitination and degradation. DCAF7 contains WD40 repeats, which facilitate specific protein-protein interactions, allowing the recognition and recruitment of substrates to the E3 ligase complex. It is particularly notable for its involvement in developmental signaling pathways, such as the JNK and Wnt pathways, where it regulates the stability of key transcription factors and signaling molecules. DCAF7 has also been implicated in craniofacial development and neural crest cell differentiation, with studies showing its interactions with kinases such as DYRK1A, which is crucial for neuronal development.
