Background

DDB1 and CUL4 associated factor 5 (DCAF5), also known as WDR82, is a substrate receptor in the CUL4-DDB1 E3 ubiquitin ligase complex and is involved in various nuclear processes, including transcriptional regulation and epigenetic modification. DCAF5 contains WD40 repeats, which enable it to interact with histone-modifying enzymes and transcription factors, mediating the ubiquitination and degradation of these nuclear substrates. It has been implicated in histone methylation, particularly by interacting with methyltransferase complexes like SETD1A/B, playing a key role in chromatin remodeling and gene expression regulation. DCAF5 is ubiquitously expressed, and its function is critical for maintaining genomic stability and orchestrating cellular responses to environmental cues. Dysregulation of DCAF5 has been linked to abnormal gene expression patterns and cancer, emphasizing its importance in both normal physiology and disease.
