Background

DDB1 and CUL4 associated factor 15 (DCAF15) serves as a key substrate receptor for the CUL4-DDB1 E3 ubiquitin ligase complex, involved in protein degradation and cellular regulation. DCAF15 is notable for its unique ability to interact with specific substrates, including RBM39, a splicing factor targeted for ubiquitin-mediated degradation by the sulfonamide family of drugs. This interaction has been extensively studied in the context of targeted protein degradation, particularly in cancer therapy. DCAF15 contains WD40 repeats that facilitate the recognition of proteins destined for ubiquitination, mediating their recruitment to the E3 ligase complex. It plays a role in regulating RNA splicing and transcription, as well as in maintaining cellular proteostasis. The expression pattern of DCAF15 across tissues and its involvement in essential cellular processes underscore its significance in both normal physiology and disease states, especially as a therapeutic target in cancers reliant on RBM39.
