DDB1 and CUL4 associated factor 12 (DCAF12) acts as a substrate receptor within the CUL4-DDB1 E3 ubiquitin ligase complex, playing a vital role in ubiquitin-mediated protein degradation. This process ensures the stability of key proteins required for various cellular functions. DCAF12 contains WD40 repeats that facilitate specific protein-protein interactions, allowing it to recognize substrates with a C-terminal diglutamate (Glu-Glu) motif and target them for ubiquitination and subsequent proteasomal degradation. One prominent example is MOV10, an RNA helicase involved in spermatogenesis and T-cell activation, whose stability is regulated by DCAF12 through recognition of a specific Glu-Leu degron. Furthermore, DCAF12 is involved in modulating apoptosis by interacting with inhibitor of apoptosis proteins (IAPs) and regulates NF-κB signaling, highlighting its importance in cell survival pathways. Its expression is ubiquitous, with particularly high levels in early spermatids, underscoring its role in male reproduction. These characteristics highlight its importance in maintaining cellular homeostasis and in specialized functions related to reproductive biology.
