Descripción
ALOX12B is a lipoxygenase enzyme that specifically oxygenates arachidonic acid to form 12(R)-hydroperoxyeicosatetraenoic acid [12(R)-HPETE], which is further metabolized to 12(R)-HETE. ALOX12B is primarily expressed in the epidermis, where it plays a critical role in skin barrier formation and maintenance. It is involved in the biosynthesis of specialized lipids required for the development of the cornified lipid envelope, a structure essential for skin permeability and protection. Mutations in ALOX12B are linked to autosomal recessive congenital ichthyosis (ARCI), a skin disorder characterized by scaling, dryness, and impaired barrier function due to defective lipid metabolism. Beyond its role in skin biology, ALOX12B has been implicated in inflammation and wound healing processes. Dysregulation of ALOX12B may contribute to skin-related disorders and inflammatory skin diseases, making it an important target for understanding epidermal homeostasis and barrier repair therapies.
