Background

ANK2, also known as ankyrin-B, is a ubiquitously expressed cytoskeletal adaptor protein that plays essential roles in the organization of membrane domains, ion channel localization, and cellular signaling. ANK2 is highly expressed in cardiac muscle and neurons, where it anchors proteins such as ion channels, transporters, and signaling molecules to the spectrin-actin cytoskeleton, ensuring the proper functioning of cellular compartments. In the heart, ANK2 regulates the localization of critical ion channels such as the Na+/K+ ATPase, Na+/Ca2+ exchanger, and voltage-gated sodium channels, making it essential for maintaining cardiac electrophysiology and calcium homeostasis. Mutations in ANK2 are associated with ankyrin-B syndrome, a disorder that includes cardiac arrhythmias, such as long QT syndrome and catecholaminergic polymorphic ventricular tachycardia (CPVT). In neurons, ANK2 is involved in axonal organization, synaptic transmission, and neurodevelopment. Dysregulation of ANK2 has been linked to neurological disorders and cardiac pathologies, underscoring its importance in maintaining ion channel function, cellular organization, and physiological homeostasis.
