ALDH1A3 is a cytosolic enzyme that catalyzes the irreversible oxidation of retinaldehyde to retinoic acid, an essential signaling molecule that regulates gene expression during development, tissue homeostasis, and differentiation. ALDH1A3 plays a pivotal role in retinoic acid synthesis, particularly in the nervous system, epithelial tissues, and metabolic organs. Its expression is highly regulated during embryogenesis, where it influences organ patterning, neurogenesis, and craniofacial development. ALDH1A3 is often overexpressed in various cancers, including glioblastoma, breast cancer, and lung cancer, where it functions as a marker for cancer stem cells (CSCs), promoting tumor initiation, chemoresistance, and metastasis. Beyond its role in cancer, ALDH1A3 is involved in lipid and glucose metabolism, and its dysregulation has been linked to metabolic disorders such as obesity and diabetes. Recent studies highlight ALDH1A3 as a potential therapeutic target for blocking CSC-mediated tumor progression and metabolic dysfunction.
