AK9 is a cytosolic and nuclear adenylate kinase isoform involved in nucleotide metabolism, cell signaling, and energy balance. AK9 catalyzes the transfer of phosphate groups between ATP/GTP and AMP, serving as a critical enzyme for maintaining ATP regeneration during cellular stress. It is highly expressed in metabolically active tissues, such as the liver, skeletal muscle, and heart, where it supports high ATP turnover and metabolic demands. AK9 has been implicated in the regulation of the cell cycle, as it ensures adequate nucleotide pools for DNA synthesis and mitosis. Mutations or dysfunction of AK9 can result in mitochondrial dysfunction, disrupted energy homeostasis, and defective cell proliferation. AK9 also interacts with signaling pathways, such as AMPK and mTOR, that regulate energy sensing and cellular growth. Studies have shown that AK9 plays an essential role in stress adaptation, apoptosis, and cell survival, particularly in hypoxia and oxidative stress conditions. AK9 deficiency has been linked to neurodegenerative disorders, cardiovascular disease, and cancer progression, where impaired energy metabolism disrupts cellular function.
