Descripción
Acylglycerol kinase (AGK) is a mitochondrial enzyme that catalyzes the phosphorylation of monoacylglycerol and diacylglycerol to produce lysophosphatidic acid (LPA) and phosphatidic acid (PA), which are critical intermediates in lipid metabolism, membrane biosynthesis, and intracellular signaling pathways. AGK also plays an essential role in the assembly and stability of mitochondrial respiratory chain supercomplexes, ensuring efficient oxidative phosphorylation and ATP production. This dual function underscores its importance in both energy metabolism and lipid signaling. Mutations in the AGK gene are associated with Sengers syndrome, a rare mitochondrial disorder characterized by cardiomyopathy, cataracts, skeletal myopathy, and lactic acidosis. Dysregulation of AGK has also been linked to cancer progression, where it enhances tumor growth by modulating lipid-derived signaling molecules and energy homeostasis. AGK is gaining attention as a therapeutic target in cancer and mitochondrial dysfunction, with research exploring its potential in restoring metabolic balance and energy efficiency.
