The NT5E gene encodes 5'-nucleotidase ecto, also known as CD73, a glycosylphosphatidylinositol (GPI)-anchored enzyme that is crucial in purinergic signaling. It catalyzes the hydrolysis of extracellular nucleotides into nucleosides, with a primary function of converting AMP (adenosine monophosphate) to adenosine. CD73 is expressed on a wide range of cell types, including endothelial cells, epithelial cells, fibroblasts, T cells, and tumor cells. It plays a fundamental role in regulating inflammation, vascular permeability, immune response, and tissue protection under stress conditions such as ischemia. Adenosine generated by CD73 is a potent signaling molecule that binds to adenosine receptors (A1, A2A, A2B, and A3) to mediate various physiological processes, including anti-inflammatory effects, vasodilation, and immunosuppression. In the immune system, CD73 suppresses pro-inflammatory responses and helps in the resolution of inflammation. This dual role makes NT5E a key molecule in both health and disease, especially in pathologies such as cancer, cardiovascular diseases, and chronic inflammatory disorders.

NT5E(5'-nucleotidase) is also named as NT5, NTE,CD73,ecto-5'-nucleotidase, and belongs to the 5'-nucleotidase family. It catalyzes the conversion at neutral pH of purine 5-prime mononucleotides to nucleosides, the preferred substrate being AMP.The enzyme occurs mainly as a dimer and the wide distribution of NT5E in the rat hippocampus, suggesting their involvement in the control of the purinergic signaling(PMID:17619139). This protein has two isoforms with the molecular weight of 63 kDa and 58 kDa and four glycosylation sites with a signalpeptide. It can exsit as a dimer(PMID:17619139). It can be detected the band of 55 kDa,64 kDa, 70 kDa, 110 kDa by western blot(PMID:17619139; 12370277; 21533188). Defects in NT5E are the cause of calcification of joints and arteries(CALJA).
