Background

HTR3E is a member of the 5-HT3 receptor family, which functions as a ligand-gated ion channel receptor. HTR3E does not form functional channels on its own but combines with HTR3A to create heteromeric receptors that fine-tune serotonin's effects on neuronal excitation. Expressed primarily in the central nervous system and gastrointestinal tract, HTR3E contributes to the modulation of emetic responses, pain signaling, and gastrointestinal motility. Its presence alters the receptor's biophysical properties, including ion conductance and pharmacological sensitivity. HTR3E polymorphisms have been associated with functional gastrointestinal disorders, such as irritable bowel syndrome (IBS), by influencing visceral hypersensitivity and serotonin-mediated gut motility. Additionally, the HTR3E subunit plays a role in chemotherapy-induced nausea and vomiting (CINV) and serves as a target for 5-HT3 antagonists like ondansetron. The interaction of HTR3E with other subunits further increases the diversity of 5-HT3 receptor functions, making it a key modulator in serotonergic neurotransmission.
