Herpes simplex virus 1 (MacIntyre strain) was cultured in BSC-1 cells and collected from both the cell and supernatant. The antigen was harvested by disrupting optimally infected monolayers in the tissue culture medium. Cellular debris was removed by centrifugation. The antigen preparation was then inactivated using gamma radiation, which primarily damages viral genetic material.
